    As therapeutic modalities continue to expand beyond small molecules and monoclonal antibodies, one principle is becoming increasingly important in drug discovery: modality should follow biology. At the recent WuXi AppTec Spring Science Forum in Cambridge, industry experts explored how researchers can build greater translational confidence as they advance emerging modalities — from ADCs and bispecific antibodies to peptides, oligonucleotides, and protein degraders.     In our latest article, we present a case study in which a client urgently required the construction of a precise three-dimensional molecular scaffold with the correct stereochemical configuration to build a biologically active molecule. The project faced several intertwined challenges, including low yield, poor cis-trans selectivity, and difficulties in scaling up the batch process. To address these challenges, WuXi AppTec’s Research Chemistry Services (RCS) team turned to photochemistry. Applying photochemistry in an industrial setting demands highly specialized equipment and expertise, requiring light energy to penetrate the reaction system uniformly and stably — a feat nearly impossible in traditional reaction vessels.     With the widespread adoption of LED technology, advances in photocatalytic methodologies, and the evolution of flow chemistry, photochemistry has transformed from a niche method into a powerful tool in modern drug discovery. For certain types of molecules, photocatalysis offers a synthetic route that may be more efficient or, in some cases, not readily achievable with conventional methods. With its mild, rapid, and heating-free characteristics, photocatalysis offers new pathways for synthesizing complex molecules such as targeted protein degraders (TPDs). By integrating high-throughput experimentation (HTE) and flow chemistry, WuXi AppTec’s chemistry platform continues to expand the scope of scalable photochemical processes. Our capabilities support client programs from early condition screening and parameter optimization through process stabilization and flow scale-up, enabling production from hundreds of grams to kilogram scale.
