Liposome generation in microfluidics often requires complex chip surface treatments that are hard to reproduce and short-lived. Mostafa Bakouei et al. (Caglar Elbuken Lab, University of Oulu & VTT) bypassed the problem with an elegant PDMS-glass capillary hybrid microfluidic chip: 🔹 Hydrophobic PDMS + hydrophilic glass capillary form W/O/W double emulsions. No coating, fully reusable.  🔹 Monodisperse with tunable size from 1.2 mm to 27 µm by swapping the capillary. Beyond the hybrid design, they explained the physics of transition from double emulsions to liposomes: 🔹 Solvent-mediated dewetting transforms double emulsions into cell-sized liposomes, while a piconewton-scale force completing the final fission. 🔹 High yield liposomes and surfactant-free formation advantageous for synthetic biology. 🩸 Today is World Sickle Cell Day. (Drépanocytose for our french public!) A few numbers to put it in perspective: - 7.7 million people are currently living with sickle cell disease worldwide - Around 515,000 babies are born with it every year - It causes an estimated 376,000 deaths annually - Nearly 90% of cases occur in low- and middle-income countries Sickle cell anemia is an inherited disorder caused by a single mutation in the β-globin gene. The result: stiff, sticky, crescent-shaped red blood cells that struggle to flow through small vessels, triggering painful vaso-occlusive crises and chronic organ damage. Because the disease is fundamentally a problem of how blood flows, microfluidics is the gold-standard tool to study it. Mimicking the geometry of arterioles and capillaries at the micron scale, it lets researchers reproduce physiological shear rates, hematocrits and pressure drops, and watch single cells behave under realistic conditions. A great example is the work of early work of Viviana Clavería, PhD, and colleagues (Frontiers in Physics, 2021), who used 50 µm glass capillaries driven by an Elveflow OB1 pressure controller to track how dense, rigid sickle red blood cells segregate from softer ones during flow. Their finding: the stiffest cells migrate toward the vessel wall, while plasma-induced aggregation suppresses this segregation, suggesting a previously underappreciated protective mechanism against vaso-occlusion.
