Most antibodies retire after one kill. This one does not. A conventional monoclonal antibody binds its target antigen, gets internalised by the cell, and is degraded along with the antigen it captured. The antibody that cleared the target is gone along with it. A sweeping antibody is engineered to escape that fate. The mechanism is pH-dependent binding. At neutral pH in the bloodstream, the antibody binds its target with high affinity. When the complex is internalised into the endosome, the pH drops. The antibody releases the antigen. FcRn — the neonatal Fc receptor — recycles the antibody back to the cell surface. The antigen is degraded. The antibody is released back into circulation and binds again. The engineering precision required is considerable. Bind too weakly at pH 7.4 and you lose efficacy. Release too slowly at pH 6.0, and the antibody cannot reset. The window between those two failure modes is where the entire therapeutic value of the format lives. For patients living with diseases driven by chronically elevated antigens — where conventional antibodies are cleared before they can make a lasting difference — a molecule that keeps working changes what treatment can look like. #EnzeneExplains #SweepingAntibodies
