Biotechnology, or genetic engineering, can be defined as the alteration of cells or biological molecules for specific applications. Biotechnology has contributed — and will contribute — to many major medical advances. To respond to the biotechnology revolution in health care, pharmacists must become familiar with this new field. With a state-of-the-art understanding of disease processes and drug effects, pharmacists will be ideally prepared to educate patients about disease states and the roles of old and new drugs in treatment plans.

Two recombinant core streptavidins were designed and characterized to understand the role of the ... more Two recombinant core streptavidins were designed and characterized to understand the role of the terminal sequences, present in naturally truncated core streptavidins, in the properties of streptavidin. One recombinant core streptavidin, Stv-25, has an amino acid sequence very similar to natural core streptavidins. The other recombinant molecule, Stv-13, has further truncation of the terminal residues and consists essentially of only the b-barrel structure characteristic of streptavidin. These recombinant core streptavidins are tetrameric and bind four biotins/molecule, as does natural streptavidin. The solubility characteristics of Stv-13, determined by varying the concentration of ammonium sulfate or ethanol, were almost the same as those of Stv-25 and natural core streptavidin. However, Stv-13 showed an enhanced structural stability compared with Stv-25 and natural core streptavidin. Evidence continues to mount that RNA plays many vital structural and functional roles of the ribo... more Evidence continues to mount that RNA plays many vital structural and functional roles of the ribosome in protein synthesis (for a review see Thompson and Hearst, 1983 a). Indeed, the recent discovery of catalytic RNA (Kruger et al., 1982; Guerrier-Takada et al., 1983) even raises the possibility that the peptidyl transferase and translocation activities of the ribosome could reside in RNA. For these reasons, it remains vitally important that the structure of the various RNAs that participate in protein synthesis be defined more completely. At present, detailed three-dimensional structure information is available for only one such RNA type, several classes of tRNA. Prospects for obtaining comparable information on the free 5S prokaryotic rRNA appear good. However, much less is known about the structures of these RNAs as they participate in protein synthesis on the ribosome. We have only secondary structure information for the larger ribosomal RNAs and for mRNAs. Most of this information is restricted to the free RNA. Studies on the ribosome-bound species are much more difficult to interpret unequivocally because many techniques really cannot distinguish well between direct information on RNA structure and information about RNA-protein contacts.

Oligonucleotides, particularly single stranded, may ultimately be of considerable use as radiopha... more Oligonucleotides, particularly single stranded, may ultimately be of considerable use as radiopharmaceuticals. We have compared a synthetic 22-base single-stranded phosphodiester DNA with its phosphorothioate analog after both were radiolabeled with 99mTc via the hydrazino nicotinamide chelator. Whole body clearance of the label in mice was much slower when introduced on the phosphorothioate (30% vs. 75% clearance at 6 hr) because of immediate and persistent accumulation in liver (47% vs. 2% injected dose/g at 4 hr). The label in both cases was present in urine primarily on low molecular weight catabolites. High-performance liquid chromatography analysis of 37 degrees C serum incubates showed serum protein binding of 99mTc in both cases (about 100% bound at 24 hr) but to different proteins. A novel class-II restriction endonuclease designated SwaI was purified from Staphylococcus warner... more A novel class-II restriction endonuclease designated SwaI was purified from Staphylococcus warneri. This enzyme cleaves adenovirus 2 DNA, SV40 DNA and M13mp7 at one site each, but does not cleave lambda, PhiX174, pBR322 or pBR328 DNA. SwaI recognizes the octanucleotide sequence 5'-ATTTAAAT-3', cleaving in the center of the recognition sequence creating blunt ended DNA fragments. SwaI was used to digest chromosomal DNA from various microorganisms and human cells.
