Available treatments provide limited kidney protection. The protective effect of the green tea polyphenol epigallocatechin-3-gallate (EGCG) in several diseases have been extensively investigated. Experimental and clinical studies have shown that the antioxidant, anti-inflammatory, and antiapoptotic properties of EGCG are promising for the treatment and/or prevention of kidney diseases. This review analyzes the available evidence on the effects, and the likely protective mechanisms of action, of EGCG in a broad spectrum of kidney diseases, including acute kidney injury, drug-induced nephrotoxicity, kidney stone disease, diabetic nephropathy, chronic kidney disease, and kidney fibrosis.

This phenomenon requires further investigation, especially in collagen-induced arthritis (CIA), as it is considered the experimental model most similar to RA. identify CIA-induced changes in NE and ACh responses in mice aortas that may suggest endothelial dysfunction in these animals. Moreover, characterize CIA-induced modifications in iNOS expression in the aortas and cardiac and renal tissues taken from these mice that may be related to possible endothelial dysfunction. Male DBA/1J mice were immunized with 100 μg of emulsified bovine collagen type II (CII) plus complete Freund's adjuvant. Twenty-one days later, these animals received a boost of an additional 100 μg plus incomplete Freund's adjuvant. Fifteen days after the onset of the disease, aortic rings from CIA and control mice were challenged with NE and ACh in an organ bath. In these animals, iNOS was detected through immunohistochemical analysis of aorta...

Mast cells (MCs) participate in the development of the inflammatory process by attracting other cell types. Methods: The MCs profile was evaluated in the endotoxin-induced uveitis model (EIU), in the acute (24 hours) and late (48 hours) phases of the inflammatory process. EIU was induced by inoculation of lipopolysaccharide, subcutaneously, into the paw of the rats. After 24 and 48 hours of the induction, the eyes were enucleated for paraffin inclusion and histopathological studies of MCs in the anterior chamber of the eye. MCs were morphologically evaluated using Toluidine blue dye, their histamine accumulation was analyzed by Alcian blue and Safranin-O method, and the presence of tryptase and chymase by immunohistochemistry. Results: The results showed larger amount of total and degranulated MCs, as well as Alcian blue and Safranin positive MCs in the acute phase of EIU, especially in the ciliary body. Few tryptase positive MCs were observed in acute and late phases of EIU, but numerous chymase positive MCs were found in the 24-hour group. The data highlight the MC modulation during EIU and indicate these cells closely linked to the inflammatory process in uveitis.

Previous studies demonstrated the antiatherogenic effects of Garcinia kola seed extracts based on their lipid lowering effects. Our recent studies showed the in vitro antioxidant and anti-inflammatory activities of the ethanolic extract of Garcinia kola stem barks (EE). For more insight on the antiatherogenic effect of EE, we investigated its activity on some key points of the atherosclerosis process. The cytotoxicity of EE as well as its effects on LDL-uptake, LPS-induced InterCellular Adhesion Molecule (ICAM-1) and Annexin-1 (Anxa-1) expression and LPS-induced DNA damage were evaluated in Human Umbilical Vein Endothelial Cells (HUVECs). EE significantly (p<0.0001) increased the cell viability in both naive and LPS-treated HUVECs. At the concentrations of 50 and 100 µg/ml, EE significantly reduced LDL-uptake by endothelial cells sti...

It is used for antidepressant and anticonvulsant purposes, as well as to treat inflammatory disorders, skin burns, abrasions, and gastric diseases, among others. Here, we investigated the biochemical, antioxidant, and histopathological effects of vervain against chronic physical stress. Male Wistar rats were submitted to chronic physical training and oral administration of 200 mg/kg of extract for 7 weeks. Control animals were not treated with either stress or vervain. Body weight was monitored during the study. Liver, kidney, spleen, testis, epididymis, heart, skeletal muscle, and brain samples were collected. Blood cholesterol, lactate dehydrogenase (LDH), bilirubin, and creatinine kinase (CREA), among others, were studied. Glutathione peroxidase (GPox) and superoxide dismutase (SOD) antioxidant activity was analyzed in the blood, liver, and kidney. Testosterone measurements were also pe...

Bisphenol A disruption promotes mammary tumor microenvironment via phenotypic cell polarization and inflammatory response

Inflammation in the established tumor microenvironment (TME) is often associated with a poor prog... more Inflammation in the established tumor microenvironment (TME) is often associated with a poor prognosis of breast cancer. Bisphenol A (BPA) is an endocrine‐disrupting chemical that acts as inflammatory promoter and tumoral facilitator in mammary tissue. Previous studies demonstrated the onset of mammary carcinogenesis at aging when BPA exposure occurred in windows of development/susceptibility. We aim to investigate the inflammatory repercussions of BPA in TME in mammary gland (MG) during neoplastic development in aging. Female Mongolian gerbils were exposed to low (50 µg/kg) or high BPA (5000 µg/kg) doses during pregnancy and lactation. They were euthanized at 18 months of age (aging) and the MG were collected for inflammatory markers and histopathological analysis. Contrarily to control MG, BPA induced carcinogenic development mediated by COX‐2 and p‐STAT3 expression. BPA was also able to promote macrophage and mast cell (MC) polarization in tumoral phenotype, evidenced by pathways...

Here we evaluated the administration of 4% NaClO in a model of clean and infected skin lesions. Methods: Lesions were obtained on the dorsal region of Wistar rats. The wounds of some animals were inoculated with Staphylococcus epidermidis (50uL, 10-7) and clean and infected lesions were treated with Trofodermin® or 4% NaClO (diluted at 0.4%) as the following groups: Trofodermin®; S. epidermidis + Trofodermin®; 4% NaClO; S. epidermidis + 4% NaClO. Wounds sizes were daily measured and the excision of lesions was performed on day 8 for histopathological evaluation and immunohistochemical studies of Annexin A1 (AnxA1), receptors for formylated peptides (FPR)1 and FPR2 and vascular endothelial growth factor (VEGF). Levels of interleukin (IL)-1β and tumor necrosis factor (TNF)-α in the supernatants of the macerated lesions were also measured. R...

Methods: EIU was induced by injection of lipopolysaccharide (LPS) into the paws of rats, which were then sacrificed after 24 and 48 h. To assess EIU in the absence of mast cells, groups of animals were pretreated with compound 48/80 (c48/80) and sacrificed after 24 h after no treatment or EIU induction. The eyes were used for histological studies and the aqueous humor (AqH) pool was used for the analysis of transmigrated cells and Anxa1 levels. In inflammatory cells, Anxa1 expression was monitored by immunohistochemistry. Results: After 24 h, rats with EIU exhibited degranulated mast cells, associated with elevated numbers of infiltrating leukocytes and the high expression of Anxa1 in the AqH and the neutrophils. After 48 h of EIU, the mast cells were intact, indicating granule re-synthesis, and there was a reduction of neutrophil transmigration and an increase in the number of ...
