Methyl-3-hydroxypyridine-5-carboxylic acid (MHPC) oxygenase (MHPCO) catalyzes the conversion of a... more Methyl-3-hydroxypyridine-5-carboxylic acid (MHPC) oxygenase (MHPCO) catalyzes the conversion of an aromatic substrate, MHPC, to an aliphatic compound,-(N-acetylaminomethylene)-succinic acid, and is involved in the degradation of vitamin B 6 by the soil bacterium Pseudomonas sp. MA-1. Using only FAD as a cofactor, MHPCO is unique in catalyzing hydroxylation and subsequent aromatic ring cleavage without requiring a metal-ion cofactor. Here, the crystallization of MHPCO is reported together with preliminary X-ray crystallographic data. An MHPCO crystal obtained by hanging-drop vapour diffusion diffracted X-rays to 2.25 A Ê resolution and belonged to the triclinic space group P1, with four molecules per asymmetric unit. crystallization communications 314 Oonanant et al. An engineered mutant V107A of the dimeric glutathione transferase enzyme from Anopheles dirus (ad... more An engineered mutant V107A of the dimeric glutathione transferase enzyme from Anopheles dirus (adgstD4-4) was cocrystallized with glutathione substrate using the hanging-drop vapour-diffusion method. The crystal diffracted to 2.47 Ã… resolution in space group P3 2 21 (unit-cell parameters a = b = 49.4, c = 272.4 Ã…). Although the crystal morphology differed from that previously obtained for the wild-type enzyme, the crystal packing was the same. At 318 K, the engineered mutant showed an enzyme stability that was increased by about 32-fold, while possessing a similar catalytic function to the wild type. Structural determination will provide valuable understanding of the role of Val107. This residue is in the dimeric interface and appears to contribute towards enhancing the physical properties of the entire protein.

Trypanosoma cruzi dihydrofolate reductase-thymidylate synthase (TcDHFR-TS) was crystallized in co... more Trypanosoma cruzi dihydrofolate reductase-thymidylate synthase (TcDHFR-TS) was crystallized in complexes with the dihydrotriazine-based or quinazoline-based antifolates C-448, cycloguanil (CYC) and Q-8 in order to gain insight into the interactions of this DHFR enzyme with classical and novel inhibitors. The TcDHFR-TS-C-448-NDP-dUMP crystal belonged to space group C222 1 with two molecules per asymmetric unit and diffracted to 2.37 Ã… resolution. The TcDHFR-TS-CYC, TcDHFR-TS-CYC-NDP and TcDHFR-TS-Q-8-NDP crystals belonged to space group P2 1 with four molecules per asymmetric unit and diffracted to 2.1, 2.6 and 2.8 Ã… resolution, respectively. Crystals belonging to the two different space groups were suitable for structure determination.

Chitinase A of Vibrio carchariae was expressed in Escherichia coli M15 host cells as a 575-amino-... more Chitinase A of Vibrio carchariae was expressed in Escherichia coli M15 host cells as a 575-amino-acid fragment with full enzymatic activity using the pQE60 expression vector. The yield of the highly purified recombinant protein was approximately 70 mg per litre of bacterial culture. The molecular mass of the expressed protein was determined by HPLC/ESI-MS to be 63 770, including the hexahistidine tag. Crystals of recombinant chitinase A were grown to a suitable size for X-ray structure analysis in a precipitant containing 10%(v/v) PEG 400, 0.1 M sodium acetate pH 4.6 and 0.125 M CaCl 2. The crystals belonged to the tetragonal space group P422, with two molecules per asymmetric unit and unitcell parameters a = b = 127.64, c = 171.42 Ã…. A complete diffraction data set was collected to 2.14 Ã… resolution using a Rigaku/MSC R-AXIS IV ++ detector system mounted on an RU-H3R rotating-anode X-ray generator.

To do so, SARS-CoV-2 is typically grown in the lab to generate viral stocks for various kinds of experimental investigations. However, accumulating evidence suggests that such viruses often undergo cell culture adaptation. Here, we systematically explored cell culture adaptation of two SARS-CoV-2 variants, namely the B.1.36.16 variant and the AY.30 variant, a sub lineage of the B.1.617.2 (Delta) variant, propagated in three different cell lines, including Vero E6, Vero E6/TMPRSS2, and Calu-3 cells.
