Tango Therapeutics is a clinical-stage biotechnology company dedicated to discovering novel drug targets and delivering the next generation of precision medicine for the treatment of cancer. Tango was launched in 2017 with a $55 million Series A investment from Third Rock Ventures. The company has established a robust product engine that leverages advances in DNA sequencing and CRISPR-based target discovery to generate breakthrough medicines that have the potential to provide deeper, more sustained benefit than today’s targeted therapies, and extend the benefit of available immuno-oncology agents. Tango Therapeutics is focused on three areas of drug development, each in well-defined patient populations currently lacking effective treatment options, and each with hallmarks of cancer that have not been targeted yet. These include: loss of tumor suppressor gene function; multiple oncogenic drivers; and immune evasion. What fuels each of Tango’s programs is an increasingly sophisticated ability to utilize synthetic lethality - the interaction between two genes that causes cell death when both are inactivated. In cancer cells, one of these genes is inactivated by mutation; the other will be inactivated by a drug. This approach leaves normal cells largely unaffected, with the potential to greatly enhance anti-tumor efficacy and reduce associated toxicity. Tango’s success will be driven by its depth of understanding of the genetic subtypes of cancer, and corresponding insights into novel drug targets and combinations uniquely relevant to each subtype. By shaping discovery efforts in this way, Tango has the potential to reach the clinic quickly, and with a clear plan for identifying the patients most likely to benefit from each new treatment, an approach that could increase both speed and probability of success in translating novel target discoveries into transformational new medicines for patients.

We are pleased to share that Phase 1/2 data from our ongoing study evaluating vopimetostat in combination with RAS(ON) inhibitors in patients with MTAP-loss, RAS-mutant pancreatic adenocarcinoma and non-small cell lung cancer has been selected for presentation at #ESMO2026. We're proud to spotlight two team members whose journeys reflect that growth at Tango isn't a straight line, but a series of curious, courageous moments. Seth Rice, Senior Manager, Clinical Scientist came to Tango expecting one role and found himself saying yes to something entirely new. That openness led him through clinical compliance and ultimately into clinical science, where he now supports first-in-human studies. Meanwhile, Katherine L., Manager, Alliance Management, joined as a scientist, followed her interest in project management, and recently stepped into alliance management where she now builds partnerships with external collaborators, expanding her strategic impact. These transitions were made possible by exceptional mentorship, encouragement and a culture that genuinely supports learning. In Seth's words, "I never had a fixed vision of what my career should look like. Saying yes changed everything." We're lucky to have them both on our team. 💜

It was a pleasure to share a look inside Tango—from our science and pipeline to what it’s like to work day-to-day at a fast-moving biotech. Students toured our labs and connected with a panel of Tango leaders and associates representing a range of roles and career paths, bringing our work to life beyond the bench. We were especially impressed by the curiosity and thoughtfulness behind their questions. We’re pleased to announce the publication of “The Discovery of TNG456: A Highly Potent, Selective, Brain‑Penetrant MTA‑Cooperative PRMT5 Inhibitor for the Treatment of MTAP‑Deleted Cancers” in the Journal of Medicinal Chemistry, led by Kevin Cottrell. This manuscript marks the first peer‑reviewed publication on TNG456, a next‑generation #PRMT5 inhibitor currently being evaluated in a Phase 1/2 clinical trial for patients with advanced or metastatic #MTAP‑deleted solid tumors, with a focus on glioblastoma. This work reflects our continued commitment to advancing PRMT5 biology and precision oncology. The data highlights the translational rationale behind TNG456 — including strong in vivo translation and comparison to vopimetostat — and demonstrates how insights across our PRMT5 portfolio continue to inform program design and clinical strategy. Excited to share that our latest article, "The Discovery of TNG456: A Highly Potent, Selective Brain-Penetrant MTA-Cooperative PRMT5 Inhibitor for the Treatment of MTAP-Deleted Cancers", has just published in the Journal of Medicinal Chemistry! TNG456 is currently in clinical studies targeting glioblastoma, an area of significant unmet need. This work represents the third compound from our MTA-cooperative PRMT5 program to advance into the clinic, joining vopimetostat (TNG462) which is currently being evaluated in pancreatic, lung, and other cancers. This paper builds on the work that led to the discoveries of both TNG908 and vopimetostat and describes the medicinal chemistry that enabled a combination of high potency and selectivity with properties amenable for blood-brain-barrier penetration. The stories of the discoveries of TNG908 and vopimetostat were also published in the Journal, as was our earlier proof-of-concept work that we started in 2017, which was the first to demonstrate that this novel MTA-cooperative mechanism is active in cells. These efforts reflect years of collaborative effort, tremendous teamwork, and important scientific contributions across disciplines. It has been a privilege to lead such a talented and collaborative team at Tango, as well as our amazing network of CRO partners, throughout these programs.
