Natural Killer (NK) cells occupy an intriguing niche between adaptive and innate immunity, and their mechanisms of action have become the focus of promising candidates for immunotherapies, with several clinical trials underway. While we have yet to FDA-approved drugs become available, the use of innovative humanized mouse models is bringing this goal closer to reality. In our new article, we explore the research, the complexity of NK cell biology, and the application of CD34+ (HSC) and PBMC-engrafted models to generate actionable data toward this end. See the data from Dr. Martin Felices for analysis of PBMC-engrafted models where B cells are targeted by a CD19-specific TriKE, as well as a CD19-expressing tumor. Their patient-specific data reveals insights in an autologous system where both tumor and PBMCs from the same patient may provide near-future potential for NK-directed therapy. Link in comments!
