What role could iron regulation play in polycythemia vera (PV)? Our investigational siRNA therapy for people living with PV is designed to silence the TMPRSS6 gene, increasing levels of hepcidin, the body's master iron regulator. This reduces the iron available to the bone marrow, with the aim of limiting red blood cell overproduction. See the science in action in our MOA video: https://bit.ly/4wN5qRW #SilenceIsGOLDen

Gene silencing works with the body to precisely target and silence disease-associated genes without permanently altering DNA. Our proprietary siRNA platform makes this precise approach possible, unlocking new treatment pathways for patients with limited options. We're pleased to share a new publication in the American Journal of Hematology. The study explores the relationship between iron overload, inflammation, and disease progression in a preclinical model of myelodysplastic syndromes (MDS). Researchers found that hepatocyte-targeted silencing of TMPRSS6 reduced iron-driven inflammation, delayed disease progression, and prolonged survival. The findings contribute to our understanding of iron homeostasis in hematologic disease and further support the potential of TMPRSS6 silencing as a therapeutic approach. Congratulations to the authors, including Silence employees Sibylle Dames, Mona Eisermann, Alberto Martinez, and Ute Schaeper, on this important contribution to the field.
