Seed quality QTLs identified in a molecular map of early maturing soybean

This study identified QTLs influencing seed quality characters in a cross of two early maturing s... more This study identified QTLs influencing seed quality characters in a cross of two early maturing soybean (Glycine max [L.] Merr.) cultivars (Ma.Belle and Proto) adapted to the short growing seasons of Central Europe. A molecular linkage map was constructed by using 113 SSR, 6 RAPD and 1 RFLP markers segregating in 82 individuals of an F 2 population. The map consists of 23 linkage groups and corresponds well to previously published soybean maps. Using phenotypic data of the F 2 -derived lines grown in five environments, four markers for protein content, three for oil content and eight for seed weight were identified. Four from fifteen seed quality QTL-regions identified in the present study were also found by other authors. Markers associated with seed weight QTLs were consistent across all environments and proved to have effects large enough to be useful in a marker-assisted breeding program, whereas protein and oil QTLs showed environmental interactions.

Development of microsatellite markers specific for the short arm of rye (Secale cereale L.) chromosome 1

We developed 74 microsatellite marker primer pairs yielding 76 polymorphic loci, specific for the... more We developed 74 microsatellite marker primer pairs yielding 76 polymorphic loci, specific for the short arm of rye chromosome 1R (1RS) in wheat background. Four libraries enriched for microsatellite motifs AG, AAG, AC and AAC were constructed from DNA of flow-sorted 1RS chromosomes and 1,290 clones were sequenced. Additionally, 2,778 BAC-end-sequences from a 1RS specific BAC library were used for microsatellite screening and marker development. From 724 designed primer pairs, 119 produced 1RS specific bands and 74 of them showed polymorphism in a set of ten rye genotypes. We show that this high attrition rate was due to the highly repetitive nature of the rye genome consisting of a large number of transposable elements. We mapped the 76 polymorphic loci physically into three regions (bins) on 1RS; 29, 30 and 17 loci were assigned to the distal, intercalary and proximal regions of the 1RS arm, respectively. The average polymorphism information content increases with distance from the centromere, which could be due to an increased recombination rate along the chromosome arm toward's the telomere. Additionally, we demonstrate, using the data of the whole rice genome, that the intra-genomic length variation of microsatellites correlates (r = 0.87) with microsatellite polymorphism. Based on these results we suggest that an analysis of the microsatellite length variation is conducted for each species prior to microsatellite development, provided that sufficient sequence information is available. This will allow to selectively design microsatellite markers for motifs likely to yield a high level of polymorphism.

Genetic diversity in camelina germplasm as revealed by seed quality characteristics and RAPD polymorphism

Camelina is an alternative oilseed crop species with limited information about the origin and div... more Camelina is an alternative oilseed crop species with limited information about the origin and diversity of available germplasm. Therefore, a set of 130 camelina accessions from a world collection was evaluated for oil content, protein content and 1000-seed weight in field experiments grown in three macro-environments in Austria. Based on phenotypic data, accessions were categorized into four groups with different seed characteristics using k-means cluster analysis or principal component extraction. Subsequently, a representative set of 41 accessions was subjected to random amplified polymorphic DNA (RAPD) analysis. Of 24 primers, 15 were polymorphic producing a total of 30 marker loci. Genetic distance estimates between the 41 accessions were calculated, based both on RAPD polymorphism and on seed quality characteristics, and dendrograms were generated for comparison. Similarities were found between the two different clustering approaches, and grouping was partly in agreement with pedigree information or geographic origin. However, as the two estimates of diversity sampled different segments of the genome, i.e. regions coding for seed characteristics or phenotypically neutral genomic regions highlighted by discrete markers, the correlation between the two distance matrices was low.
