The analysis of the polymorphism sequence hypervariable region of mtDNA sequence polymorphisms ha... more The analysis of the polymorphism sequence hypervariable region of mtDNA sequence polymorphisms has had a profound impact on understanding identification of the wild ancestral species, events surrounding the domestication and the establishment of the geographical distribution of genetic diversity. We observed and tested a new test-system of analysis of the polymorphism of mtDNA D-loop region, and universal for the species of Sus Scrofa. We have analyzed a variability of mtDNA and nucleotide sequence variation in mitochondrial DNA by sequencing the D-loop region in domestic pigs (large-white breed n=50, landras n=20). Obtained sequences had been compared between themselves and with the sequences from the NCBI gene Bank. The dendrogram analysis indicates that were revealed special clusters between different breeds. Polymorphism data analysis of mitochondrial DNA of domestic pigs had revealed polymorphic positions and variability of mtDNA and nucleotide sequence variation in mitochondrial DNA between different breeds. Polymorphism data analysis of mitochondrial DNA of domestic pigs in our study contained essential polymorphic positions: transition and transversion elements and it could be used for detection of mother inheritance. In this study, we describe transition and transversion, indels and we didn't find a deletion, in the positions from 15797 to 16797 was found a transversion A → G. Wild boar (Sus scrofa) distributed across many countries of Europe, Asia, and Africa belongs to the Suidae family [1]. Genetic variability and the population structure of the European populations are not well-known, in particular in the Russia. Our aim was to identify and evaluate mitochondrial diversity of wild boars in the Russia. We observed the variation and polymorphism in mitochondrial DNA by sequencing the D-loop region in wild boars of Russian population. A D-loop fragment of approximately 1770 bp was amplified by PCR. Sequencing of DNA amplicons identified haplotypes previously described as European and Asian types. The mtDNA sequences of wild boars (110 n) were found to be distributed in haplotypes. Obtained sequences had been compared between themselves and with the sequences from the NCBI gene Bank. Polymorphism data analysis of mitochondrial DNA of domestic pigs had revealed polymorphic positions of SNP’s. Russian wild boars had European (14n) and Asian (35n) haplotypes. We studied populations showed moderate haplotype (gene) diversity, Hd ± SD (0,961 ± 0,010) and nucleotide diversity, π ± SD (0, 02549 ± 0, 00330). The group of population of wild boar from 14 regions of Russia (Kirov region, Krasnodar region, Tyumen region, Udmurtia, Nizhny Novgorod region, Volgograd region, Vladimir region, Smolensk, Arhangelsk region, Khabarovsk region, Kurgan region, Primorsky Krai, Omsk region, Chelyabinsk), and sample of wild boar from Spain, Extremadura.

All-Russian Research Institute of Animal Breeding of the Russian Academy of Agricultural Sciences... more All-Russian Research Institute of Animal Breeding of the Russian Academy of Agricultural Sciences 142132, Moscow region, Podolsky district, village of Dubrovitsy, H.60 IGF-2 (insulin like growth factor-2) is of interest as a genetic marker because of its chromosomal location, paternal expression effects on myogenesis and participation in a wide variety of metabolic processes, and differentiating. IGF2 gene polymorphism were investigated at pigs, Russian, Belarusian, Canadian and European ancestry. High frequencies of allele A and AA genotype characterized by commercial breeds and crosses. Particularly high frequency of the desired allele A (70 %-100%) were observed the Duroc European and Canadian origin and Yorkshire breeds of swine. Low values of A allele and AA genotype in Russian and Belarusian pigs are probably the result of preferential selection of animals and lack of prolificacy BLUP evaluation in animal breeding. During the study of economically valuable traits of pigs and their offspring based on IGF2 gene variants have shown that individuals with the AA genotype and their descendants was significantly lower backfat thickness (up to 2-7 mm) above the growth rate (for 6-19 days) and less than the cost of feed (on k.ed. 0,14-0,25). These animals had a large area of eye muscle (differences amounted to 12.5 sm.kv.). An evaluation of the productivity of more than 5,000 offspring obtained from fathers with various genotypes of this marker allowed per 1,000 head of commercial pigs get 8.7 tonnes of live weight and 6.1 ton carcasses more. IGF2 genetic marker of interest in breeding farms and Russia on IGF2 genotype was introduced as an additional criterion for the selection of repair boars.
