Keep up the good work!"

• Caco-2 is not outdated in many cases. But the way we use it may need an upgrade. A nice paper by Dong Ye et al. shows why permeability prediction becomes difficult when we move beyond “easy” small molecules. For poorly soluble and/or poorly permeable compounds, standard static Caco-2 assays can struggle to provide realistic readouts. The authors explored a mucin-protected Caco-2 assay combined with complex biorelevant media, including #TIM-derived #IntestinalFluids, to better study #DrugPermeation under physiologically relevant conditions. A very relevant point: #ApparentPermeability is not only about the cell layer. It is also shaped by the assay environment: mucus interactions, medium composition, prandial state, solubility limitations, and even the unstirred water layer. This is exactly where we believe the next generation of #GutPermeability models should focus: → keeping the scalability and familiarity of Transwell-based Caco-2 workflows → adding more physiological conditions, such as mucus and biorelevant intestinal fluids → using controlled flow to reduce static artefacts and better mimic the intestinal environment → improving robustness for difficult-to-assess molecules, including low-solubility and/or low-permeability compounds At ARTIC, we are developing dynamic Transwell-based gut models designed to bring physiological flow into standard lab workflows — without adding unnecessary operational complexity. • #Bioavailability prediction remains a challenge. A nice comparative study by Moyer et al. looked at Caco-2, primary human enteroid-derived cells, and #MPS models for intestinal permeability and oral bioavailability prediction. An important take-away: more complex or more physiologically advanced models do not automatically lead to better predictive performance. Context of use matters. For highly permeable small molecules, conventional Caco-2 can still perform very well. But the bigger question is: what happens when permeability becomes the actual bottleneck? That is where many teams run into challenges with low-permeability and/or low-solubility molecules, especially when they want to combine physiological relevance with scalable, reproducible, industry-compatible workflows. Are you working on difficult-to-assess molecules in gut permeability or ADME screening? Let’s connect. At ARTIC, this is exactly the space we are working in: dynamic gut permeability models that add controlled flow while staying close to standard Transwell-based lab workflows. Contact our scientific lead Hossein Amirabadi (info@artic.technology) or send us a message on LinkedIn to have a chat. Link to the paper: https://lnkd.in/ewMyAq-N.
